The United States Pharmacopoeia standard has a clear limitation on the peroxide impurity of Fulvestrant , which requires less than 0.2% of the peroxide impurity. Therefore, how to control the oxidation of impurities in the last step is the key solve issues among the production of many producers. The development of new oxidative conditions, the selective oxidation of fulvestrant N-1 CAS number 153004-31-0 is of particular importance. It has been found that β-cyclodextrin / NBS (N-bromosuccinimide) as oxidant can not only oxidize fulvestrantN-1 to fulvestrant, but also does not produce peroxidic impurities. This process conditions are mild and the reagents used are a mixed solvent of water and acetone. after obtaining the crude fulvestrant, then recrystallization from a mixed solvent of ethyl acetate / methyl tert-butyl ether to get high purity fulvestrant, So this process has obvious advantages in the large-scale preparation.
