Product Description

Methyl 4-chloroacetoacetate CAS number is 32807-28-6, It used diketene as a starting material, obtained by chlorination, ring opening, and esterification. It is a common carbonyl compound. It is mainly used for the synthesis of Sitafloxacin ternary compound and oxiracetam. In the synthesis process of Oxiracetam, Use Methyl 4-chloroacetoacetate cas 32807-28-6 as the starting material, and the whole process can be completed in two steps:
(1) Put Methyl 4-chloroacetoacetate and metal catalyst into an autoclave, add alcohol solvent and an appropriate amount of achiral phosphine ligand, control the temperature between 50-100°C, and replace with nitrogen at a pressure of 5 atm three times, and then replaced with hydrogen pressure of 5atm three times, and then process hydrogenation reaction, the required hydrogen pressure of 10-100atm, the reaction until the hydrogen pressure is no longer reduced, get 4-chloro-3-hydroxybutyrate;
(2) The 4-chloro-3-hydroxybutyrate obtained in step (1) is added to an appropriate amount of an inorganic base and glycine amide hydrochloride, and cyclization is conducted to obtain the product oxiracetam.
In the synthesis of Sitafloxacin ternary compound, 1 kg of 4-chloroacetoacetic acid is dissolved with 5-6 L of acetonitrile, and 38-40 g of proline is added, followed by 1.25kg 1,2-dibromoethane, add 2.3-2.5kg of potassium carbonate in 8-10 batches under stirring, then control the temperature of the reaction solution at 30-36°C, stir for 30-36 hours, filter, and concentrate the filtrate under reduced pressure to dryness to obtain the Sitafloxac internary compound compound.
