Flibanserin intermediate named 1,3-Dihydro-1-(1-methylethenyl)-2H-benzimidazole-2-one CAS number is 52099-72-6, molecular formula of C10H10N2O, molecular weight of 174.2. The related literature describes his synthesis process, starting with benzoylacetate and ring-closing reaction with o-phenylenediamine to obtain the target compound 1,3-Dihydro-1-(1-methylethenyl)-2H-benzimidazole-2-one CAS number 52099-72-6. The specific process is: adding 1L of ethanol to the reaction flask, adding 150g of o-diphenylamine, adding 300g of potassium hydroxide, then adding 172g of ethyl acetoacetate dropwise, stirring at room temperature, and controlling the reaction; after the reaction is finished, the reaction liquid is concentrated. The reaction solution was added to water, filtered, washed with water, and dried to obtain 241 g of 1,3-dihydro-1-(1-methylethynyl)-2H-benzimidazol-2-one in a yield of 91%;
Secondly, 10 g of 1-isopropenyl-2-benzimidazolone was added to 50 mL of DMF, cooled to 10℃ in an ice bath, and the temperature was controlled below 20℃. 4 g of potassium carbonate was slowly added, and the mixture was stirred at room temperature for 1 hour. After the dropwise addition, 14.2 g of 2-bromoethanol was added dropwise, and the mixture was stirred at room temperature for 8 hours. The reaction mixture was poured into ice water and extracted with dichloromethane. The extract was washed with water, dried, and evaporated to dryness (1-Methylethynyl)-2H-benzimidazol-2-one, yield is 80%.
