Product Description

Baritinib intermediate is 1-(1-ethoxyethyl)-4-(4,4,5,5-tetramethyl-1,3,2-dioxaborolan-2-yl)-1H-pyrazole CAS number 1029716-44-6. There are two main synthetic methods, the first one is use 4-bromo-1-(1-ethoxyethyl)-1H-pyrazole and 2-methoxy-4,4,5,5-four Methyl-1,3,2-dioxaborolan as the starting material, and the target product is obtained by one-step reaction. The yield of this method is only 50%. The second type: 4-bromo-1-(1-ethoxyethyl)-1H-pyrazole and boranoic acid pinacol ester are used as raw materials to obtain the target product through one-step reaction. The yield of this method is only 65. %. The yields of the above two methods are not ideal.

    One method is ideal, in the presence catalyst of 0.1 equivalent of Boron trifluoride etherate, 1 equivalent of 2-chloromalonaldehyde is added to the toluene solvent and heated to 80-120℃, and the reaction is carried out to obtain 4-chloropyrazole, spin dry After the solvent, anhydrous ethanol, 2.0 equivalents of tetrahydroxydiboron, 3.0 equivalents of diisopropylethylamine, 0.05 equivalents of NiCl2 and 0.10 equivalents of PPh3 are directly added, followed by reaction at 20-80℃for 4-16 hours, and the inorganic particles are filtered off by cooling. The salt and the filtrate are evaporated to dryness. After adding ethyl acetate, the mixture is washed with water, and the organic layer is added with 1.2-1.5 equivalents of pinacol.
