Product Description

Dabigatran etexilate intermediate with the chemical name of 3-(Pyridin-2-ylaMino)-propionic acid ethyl ester CAS number 103041-38-9,Exactly speaking,it is the side chain of the intermediate of dabigatran etexilate in the fifth step. The appearance is off-white to white powder. Its usually produces the fifth step intermediate of dabigatran etexilate by reaction with 3-bromo-4- (methylamino) benzoic acid,details as follows: 34.4 g of 3-bromo-4- (methylamino),mixed with 240 mL of dichloromethane at room temperature and stirred for 1 hour and cooled to -20 ° C. 12.0 mL of thionyl chloride was added and then about 0.5 mL of N, N-dimethylformamide was added dropwise to continue the reaction for 4 hours. Until naturally rising to room temperature, then 29.12gram of ethyl 3- (pyridin-2-ylamino) propionate , 41.7ml were added, continued reaction for 6 hours at room temperature, the reaction was terminated by adding 300 mL of water, the organic layer was separated, The mixture was further washed with 200 mL of saturated aqueous sodium chloride solution, dried over anhydrous sodium sulfate and concentrated under reduced pressure to a total volume of about 100 mL. 50 mL of n-hexane was added to precipitate a solid which was stirred for 2.5 hours at room temperature. Vacumn Dried for 1 hour to give a white solid 57.7 g, this white solid was dabigatran etexilate intermediate N-5.
