Alectinib intermediate 2 has the chemical name of tert-butyl 6-cyano-2-(2-(4-ethyl-3-iodophenyl)propan-2-yl)-1H-indole-3-carboxylate, cas number is1256584-75-4,use mono-tert-butyl malonate and 3-iodo-4-ethyl-tert-butylbenzene as the starting material, through 
Condensation, cyclization, condensation with 4-(4-piperidinyl)morpholine , mixed tert-Butyl (4-ethyl-3-iodophenyl)-4-methyl-3-oxopentanoate (6.3 g, 15 mmol) with 3-cyanobenzoquinone (2.5 g, 19 mmol) and trifluoroacetic acid (94.9 g, 832 mmol) was mixed, the reaction mixture was stirred at 100 ℃ for 9 hours, the reaction liquid was lowered to 60℃, 50ml water was added, the reaction was kept warm for 3 hours, and the mixture was evaporated to dryness. The mixture was extracted with ethyl acetate. Drying over magnesium sulfate, concentrating to dryness by rotary evaporation, and recrystallized from ethyl acetate and n-hexane to obtain off-white solid 6.2 g tert-butyl 6-cyano-2-(2-(4-ethyl-3-iodophenyl)propan-2-yl)-1H-indole-3-carboxylate, cas number 1256584-75-4,yield is 90%.
