Product Description

4-Nitro LenalidoMide CAS number is 827026-45-9, which is the N-1 intermediate of lenalidomide. His preparation method is as follows: Firstly, under ice bath, add 80 ml of concentrated nitric acid to 450 ml of 98% concentrated sulfuric acid, stir for 10 min, continue to keep the ice bath, add iso-pyroline 50 g under vigorous stirring, stir for 30 min, continue stirring for 8 hours at room temperature. The ice water was quenched, suction filtered, and the filter cake was a crude product. The crude product was washed successively, washed with saturated sodium hydrogen carbonate and then recrystallized from petroleum ether to give 58.3 g of pale yellow solid 4-nitroisoporphyrin.
In the second step, 16.4 g of 4-nitroisoindoline, 21.1 g of 3-bromo-2,6-piperidinedione, 5.7 g of cuprous iodide, 53 g of sodium carbonate were added to the flask, and 200 ml of acetonitrile was added as a reaction. The solvent is heated to 55℃, and the reaction is stirred for 5 hours. The reaction mixture is concentrated, poured into water, and extracted with dichloromethane. The organic phase is washed with water, washed with saturated salt solution, dried over anhydrous sodium sulfate and evaporated. Recrystallization with 1:10 dichloromethane:N-hexane solution to give the desired compound 24.4 g 4-Nitro LenalidoMide CAS number 827026-45-9.
